Overall Format and structure:
I like that the article as a whole covers many different perspectives of DNA synthesis and assembly. The processes are explained and pieces about the history and economics of their use are woven in seamlessly.
Introduction and background material:
The introduction is a little informal and editorialized. It should probably be entirely rewritten.
Methods and main body/concepts:
While the article covers a lot of information, it's pretty reliant on links to other websites to deliver some of the information that is only hinted at in the body. Possibly the scope of the article could be limited and more fleshed out within that scope.
Relation to iGEM and future directions:
There's no mention of iGEM or future directions. BioBricks and the Registry are mentioned, but not in the context of the competition or their use for projects. I think one way to bring iGEM in would be to talk about how different assembly methods are used in various iGEM projects.
Figures, Figure legends, and citations:
I feel that the figures could be annotated better to help explain the processes. Additionally, there could be a table comparing different assembly processes, as is common in many discussions of assembly techniques. Some of the figures have citations and some do not. I believe that most of the schematic figures without citations are original creations, but should still be cited as such in their description. Generally, the citations in the article are good and there are even descriptions in the list of references at the end, which is ideal.
